K. pneumoniae NK-5 (1 × 107 cells/ml) was cultured with various concentrations of ConA in TSB, and bacterial growth was determined by counting colonies on LB agar plates, as described in Materials and Methods. One of three experiments is represented. Amount of ConA: ⋄, none; ▪, 1 μg/ml; ▵, 10 μg/ml; •, 50 μg/ml. The binding (B) and agglutinating (C) activity of ConA for K. pneumoniae. K. pneumoniae cells (5 × 107/ml) were coincubated with 20 μg/ml of ConA, 20 μg/ml of fluorescein-conjugated ConA, or 20 μg/ml of fluorescein-conjugated ConA plus 10 μg/ml of ConA in PBS as described in Materials and Methods, and the binding activity was determined by flow cytometric analysis.
