Pancreatic islets were isolated from wild-type (black bars) or iPLA2β-null (cross-hatched bars) mice that had been fed a high fat diet and incubated (30 islets/condition, 30 min, 37 °C) at a glucose concentration of 3, 8, or 20 mM in the absence or presence of 2.5 μM forskolin. Medium was then removed for measurement of insulin by radioimmunoassay. Values represent mean ± S.E. (n = 7). Single asterisk denotes p < 0.01, and double asterisk denotes p < 0.001 versus wild-type.
