Quantitative analysis of Ad vector DNA accumulated after transduction of CAR-negative and CAR-positive cells. Human H1299 and MCF-7 cells were exposed for 2 h to 10,000 vp/cell of Ad5-EGFP, CELO-EGFP, and CELOEGFP-HIRGD. The virus-containing medium was then removed, and the cells were washed twice with 0.5 ml of ice-cold phosphate-buffered saline and incubated in fresh medium for 48 h. Total DNA was extracted and used in real-time PCR to determine the number of Ad copies present in the cell population. The error bars represent standard deviations from the mean.
