Procyclic trypomastigotes [non-induced (A and C, continuous line) or after 4 days of induction with tetracycline (B and C, broken line); 0.1 mg of protein/ml] were added to a medium containing 125 mM sucrose, 65 mM KCl, 2 mM MgCl₂, 10 mM Hepes (pH 7.4), 1.5 μM digitonin and 3 μM Acridine Orange. ATP (1 mM), PP_i (0.2 mM), nigericin (1 μM) or NH₄Cl (10 mM) was added where indicated. The scale for (A) and (B) is the same and is shown under (B). The results shown are representative of at least three experiments with different cell preparations.
