(A) Crosslinking in the absence (lane 1) and presence (lane 2) of 10−5 M [Nle8,18,Tyr34]bPTH-(1–34)NH2. The arrow shows the position of the predominant ≈87-kDa crosslinked ligand–receptor conjugate. (B) SDS/PAGE in the absence (lane 1) and presence (lane 2) of 2-mercaptoethanol. Deglycosylation of the initial ligand–receptor conjugate was achieved by treatment with Endo F (lane 3). Arrows indicate the positions of the photocrosslinked receptor (≈87 kDa) and its deglycosylated form (≈59 kDa). Samples were loaded on 7.5% (wt/vol) SDS/PAGE. Molecular mass markers are also indicated.
