Cell lysates were prepared from HeLa cells transfected with pCMV-His-pXBP1(S) and/or pCMV-pXBP1(U), and the lysates were subjected to pull-down assays using Ni-NTA agarose. Input and bound materials were analyzed by immunoblotting with anti–XBP1-A antibody. (B) Mixing of two proteins synthesized in vitro. Indicated proteins were translated in vitro using rabbit reticulocyte lysates and analyzed essentially as in A. *, nonspecific banding.
