Truncated milton can localize to mitochondria. COS7 cells were transfected with either COOH-terminal regions of milton-A (amino acids 750–1,116 and 847–1,116, and also carrying a flag epitope tag) or NH2-terminal regions (amino acids 1–450 and 1–750). Cells were labeled with MitoTracker orange and anti-milton mAb 5A124 (A and B) or 2A108 (C and D). Constructs containing the region 847–1,116 were localized to mitochondria, but those that lacked this region were not. Bar, 10 μm.
