(A) Transactivation of PCNA through CREB and CBP by E1A requires p300/CBP binding regions of E1A 243R. The G5PCNA-CAT reporter plasmid was cotransfected into HeLa cells with either GAL4-CREB or GAL4-CBP and wild-type, or pCMV12S.FS, or mutant E1A expression plasmids (pCMVΔ2–36, pCMVΔ30–85, pCMVΔ2–85, pCMVΔ86–120, and pCMVΔ120–140) as indicated. CAT activity values represent an average of three independent experiments performed in duplicate with SD noted. (B) Transactivation of CREB by E1A requires CBP. HeLa cells were transiently transfected with the G5PCNA-CAT reporter plasmid and either GAL4-CREB or GAL4-CREB pm and wild-type pCMV12S or the control pCMV12S.FS plasmid.
