Fura-2-loaded HeLa cells incubated in 1 mm Ca2+-containing standard medium were stimulated with histamine and treated with PPT as indicated. AC, show single-cell Ca2+ records from three different experiments. Traces of four representative single cells present in the same microscope field for each experiment are shown. The bottom traces show the mean of all the cells analysed in each experiment (20, 29 and 31 cells for A, B and C, respectively; note the enhanced scale). The initial histamine-stimulated [Ca2+]c peaks are truncated for convenience. Data are representative of 412 analysed cells.
