Figure 5. CO restores mitochondrial antioxidant enzyme and cardiac cGMP content in DOX-treated mice.

Figure 5

(A) Western blots of cardiac HO-1 (compared with tubulin) and mitochondrial SOD and glutathione peroxidase (mtGPX) relative to porin. (BP) Fluorescence microscopy localizing SOD2 (red) to the mitochondria (green). Control LV showed intense SOD2 labeling in cardiomyocyte mitochondria (BD); CO increased mitochondrial staining intensity (EG); sections of LV in DOX-treated mice after 14 days (HJ) showed scattered SOD2 labeling in mitochondria; LV of mice treated with DOX plus 1 dose of CO (KM) and LV of mice treated with DOX plus 2 doses of CO (NP) were improved. The histogram (Q) demonstrates loss of cGMP in myocardium after DOX was restored by CO. Values are means ± SEM of 6 experiments. *P < 0.05 versus other groups. Original magnification, ×300.
