Short-term incubations with histamine suppress IL-12 production. iDCs were incubated with LPS in the presence or absence of histamine for the indicated periods of time, after which the cells were washed and replated with fresh medium containing LPS but no histamine. Secreted IL-12 was determined after a total of 24 hours. Similar results were obtained in two other experiments. The histamine-induced decrease was significant (P < 0.01) at all timepoints. P values for DCs treated with LPS plus histamine: 0.12 (1 hour vs. 3 hours); < 0.001 (1 hour vs. 6 hours); and 0.04 (1 hour vs. 24 hours).
