Nuclear extracts were prepared and the p65 subunit of NF-κB was measured by western blot. (b) Effects of PPARα agonists on STAT1α activity in J774 macrophages. Cells were stimulated by LPS (10 ng ml⁻¹) and treated with WY14643 (100 μM) for 6 h. Nuclear extracts were prepared and STAT1α was measured by western blot. Protein levels are expressed relative to that in LPS-treated cells (set to 100%). Lamin A was used as a loading control. Results are expressed as mean±s.e.m. (n=3).
