Concentration-dependent inhibition of NTPDase1 by reactive blue 2 (▪), ARL67156 (▾) suramin (•) and PPADS (dM) determined by capillary electrophoresis using in-capillary reaction at capillary inlet, using a substrate concentration of 320 µM ATP, a reaction buffer consisting of 140 mM NaCl, 5 mM KCl, 1 mM MgCl2, 2 mM CaCl2, and 10 mM Hepes, pH 7.4, and various concentrations of inhibitor. The separation conditions were 50 mM phosphate buffer, pH 6.5, neutral capillary, 30 cm length (20 cm to the detector), 50 µM I.D.; −60 µA, 7 kV; capillary cartridge temperature 37 °C; detection at 210 nm, pressure injection. Data points represent means T SD from three separate experiments, each run in duplicate.
