Functional complementation of FtsY depleted cells by LacY–FtsY hybrids. E. coli (N4156::pAra14-FtsY′) cells harboring the indicated plasmids were grown overnight in LB with arabinose, washed once in LB, and diluted to OD₆₀₀ = 0.005 in LB with or without arabinose as indicated. Growth curves (A) were constructed from the average of two independent experiments. After 4 hr, 10 ml samples were transferred from chosen cultures into separate Erlenmeyers and induced by 0.5 mM IPTG for 2 hr. Membranes were prepared from the induced cells and subjected to Western blot analysis using anti-LacY antibodies.
