Cells were incubated with or without HA or methyl cellulose and metabolically labeled with [³⁵S]-sulfate as described in Materials and Methods. Culture media were collected and cell layers extracted and purified using DEAE chromatography. Aliquots were then digested with chondroitin ABC-lyase or heparitinase and analyzed on Sepharose CL 4B columns. Results are expressed as ³⁵S-sulfate cpm/10³cells ± 1 SD (n = 5 for LMW and HMW HA and n = 3 for methyl cellulose).
