Effector cytokine production is reduced in CD40KO mice. Splenocytes from C57BL/6 and CD40KO mice primed with WT DCs and boosted with peptide-CFA were activated in vitro with peptide (A) or PMA/ionomycin (B and C). In A, the IFN-γ–producing cells were detected by cell surface cytokine capture assay and in B and C by intracellular cytokine staining. The percentages of total CD4 cells producing either IFN-γ (A and C) or IL-2 (B) are shown. Each point represents one mouse, and the line shows the mean of each group.
