Impaired TLR-mediated cytokine and chemokine production in macrophages from kinase–inactive IRAK4 knock-in mice. (a) Wild-type (WT), IRAK4 kinase–inactive knock-in (KI), or IRAK4 knock-out (KO) macrophages were treated with TLR ligands LPS (1 μg ml−1), R848 (1 μg ml−1), or CpG ODN (4 μg ml−1) for 24 h. IL-6, KC, and TNF-α concentrations were measured by ELISA. Results shown are the means (± SD) of triplicate determinations. (b) WT or KI macrophages were treated with IL-1β (10 ng ml−1) for 24 h.
