Accumulation of open coding ends in Lig4−/− p53p/p thymocytes. (A) Diagram of V(D)J recombination activity at the TCRδ locus. Cleavage by Rag nucleases generates a blunt signal end (se) and hairpin coding end (ce), which can be further processed by the DNA–PK–Artemis complex. (B) LMPCR assay of WT and Lig4−/− p53p/p (LP) thymocytes, signal ends can be digested by ApaLI to reduce the PCR products from 123 to 98 bp.
