Agonist-induced arachidonic acid release from Sf9 cells expressing wild-type or phosphorylation-site mutants of cPLA₂. [³H]Arachidonic acid release from Sf9 cells was measured after treatment with A23187 (2 μg/ml) or okadaic acid (1 μM) for 90 min, or with CryIC toxin (2 μg/ml) for 20 min. The amount of label released into the medium is expressed as a percentage of the total radioactivity incorporated (cell-associated plus medium). Data represent the mean ± SEM of three experiments performed in triplicate. The bottom panel shows a representative immunoblot of cPLA₂.
