185-kD proEGF is preferentially cleaved from the basolateral cell surface in MDCK cells. (A) MDCK cells transfected with proEGF were grown on Transwell filters and labeled for 2 h with ³⁵S-Translabel. Cells were then washed and chased for time periods indicated. After the chase, cell surface (data not shown) and media (A) immunoprecipitations were performed using mAb-EGF. (B) Quantitation of soluble 170-kD EGF released into apical (open circles) and basolateral (closed circles) medium. Average of two separate experiments ± SD are shown and expressed in arbitrary units calculated from total soluble 170-kD EGF released at each time point.
