Nucleotide and predicted amino acid sequence of the rolling stone transcription unit. Three different overlapping cDNAs encompassing 1,212 bp were selected for sequencing. The longest cDNA that we have isolated so far did not contain the putative start codon. Translated into an amino acid, this cDNA is missing the first two amino acids shown in the figure. Therefore, we completed the open reading frame by sequencing genomic clones using synthetic oligonucleotides. The genomic sequence (from nucleotide 1 to 399) shown in this figure begins with a SacI restriction site. This site is identical to the SacI site left from the P-element insertion site shown in Fig. 1 A. The sequence between nucleotide 400 and 1612 is based on cDNAs. The first methionine of the deduced Rost amino acid sequence is numbered 1. The positions of the three introns were marked with symbols (T). The P-element insertion is labeled with an asterisk.
