Effect of antisense oligonucleotides on the polarity of sucrase isomaltase in CACO-2 cells. CACO- 2 C2BBe1 monolayers were grown and incubated with random (a), random/2 (c), A19 (b), or A19/2 (d) oligonucleotides as described before. The cells were fixed in 3% PFA and processed for indirect immunofluorescence with a mAb anti-sucrase isomaltase. The monolayers were analyzed under a confocal microscope, and three-dimensional reconstructions in a plane perpendicular to the monolayer were obtained from stacks of confocal optical sections.
