Porcine sperm cells (2 × 106 cells per ml) and oocytes (∼500 oocytes per ml) were resuspended in HBSS/BSA (−), or calcium and magnesium–free HBSS containing 2 mM EDTA (EDTA). The sperm cells and oocytes were mixed in the presence of A23187 unless specifically indicated. Aliquots of oocytes were preincubated separately with mouse IgM, CSLEX (an mAb against SLex), PL5 (an mAb against PSGL-1; all at 30 μg/ml), or platelet P-selectin (10 μg/ml), or human IgG, P-, and E-selectin Rgs (all at 30 μg/ml), respectively. Aliquots of sperm cells were preincubated with mouse IgG, P7 (a leukocyte adhesion blocking mAb against P-selectin), or P23 (a leukocyte adhesion nonblocking mAb against P-selectin; all at 30 μg/ml) at 22°C for 30 min. All results were expressed as the mean ± SD number of adherent sperm cells per oocyte in the optical plane studied in five separate experiments; more than 50 oocytes were observed for each condition.
