Dynein-GFP accumulation at the SPB. This sequence of fluorescence stacks demonstrates the accumulation of dynein on separated SPBs in budded cells. Cell cycle progression is from left to right at 5-min intervals. The upper left cell has only one visible SPB by fluorescence. In 5 min, the second SPB can be visualized, well separated from the first (∼1.4 μm). Over time, the second SPB acquired fluorescence equal in intensity to the first. Quantitation of fluorescence accumulation over time is graphed in the panel to the right. Bar, 2 μm.
