Neither the V or C domain mutants are transport competent. COS-1 cells were transfected with the cDNA clones for WTλ, V_mutλ, and C_mutλ. Cells were metabolically labeled for 3 h. Culture supernatants were saved and immunoprecipitated with anti-λ (λ_m), and lysates were prepared, divided, and then immunoprecipitated with either anti-λ followed by protein A–Sepharose (λ) or protein A–Sepharose (P.A.) alone (to control for nonspecific binding). Samples were analyzed on SDS gels under reducing conditions.
