Yeast cells were pre-embedding labeled with anti–protein A antibody conjugated to 8-nm colloidal gold (see Materials and Methods). The mean ± SD are given from all values. 22, 15, and 19 cells were analyzed for the wild-type (Wt), ProtA-DHFR, and ProtA-Pus1p strains, respectively. The quantitation was performed by a stereological counting method (Lucocq, 1992).
