Cells were incubated with C6-NBD-GlcCer or -SM at 37°C for 30 min. The fluorescent pool of C6-NBD-lipids in the basolateral PM was then back exchanged in the absence or presence of nocodazole. Cells were then rewarmed to and further incubated in back exchange medium with or without nocodazole at 18°C for 1 h. The percentage of NBD-positive BC (i.e., BC alone and BC + SAC) was determined as described in Materials and Methods. The percentage of NBD-positive BC before the chase was set at 100%. Data are expressed as mean ± SEM of at least two independent experiments.
