Effect of 2 h stimulation on nuclear translocation of NFATc. (A) Two nuclei from a fiber expressing NFATc–GFP and stimulated with 5-s trains of 10 Hz stimuli every 50 s for 2 h. Duration of stimulation is labeled above the images. (B) Time course of nuclear translocation of NFATc in fibers stimulated with 5-s trains for up to 2 h. Data are presented as entire nuclear fluorescence (open circles), intranuclear foci fluorescence (filled circles), and cytoplasmic fluorescence (open squares). Stimulation resulted in increasing fluorescence in nuclei and formation of intranuclear foci during the first 30–40 min stimulation. Further stimulation did not cause noticeable increase of fluorescence, as seen in both A and B. Results are expressed as the mean ±SE. (B) 28 foci of 10 nuclei from 6 fibers. Bar, 10 μm.
