Identification of a region in the cytoplasmic domain of P0 that is essential for P0-mediated adhesion. (A) L cells stably expressing full-length or truncated P0 were labeled with a vital fluorescent dye and assayed for adhesion to monolayers of L cells expressing full-length P0. Adherent cells were determined by measuring the amount of fluorescence associated with the cell layers. Adhesion of L cells bearing full-length P0 was considered 100% and used to calculate adhesion of the other lines. Each cell type was assayed in triplicate. (B) HeLa cells stably expressing full-length or truncated P0 were assayed for adhesion as described in A. WT, full-length P0; Δ13, Δ28, Δ43, Δ59, P0 bearing deletions of the terminal 13, 28, 43, and 59 amino acids, respectively; Vect, L cells transfected with empty vector.
