ER-to-Golgi transport is required for degradation of soluble but not membrane-bound ERAD substrates. (A–D) Wild-type and ER transport mutant strains sec12–4 and sec18–1 expressing HA-tagged ERAD substrates were grown to log phase at 22°C and shifted to the restrictive temperature of 37°C for 30 min. Time courses were performed and analyzed as described in the legend to Fig. 2. The data is plotted to compare rates of degradation for each substrate in various strain backgrounds. A Δcue1 strain was included as a positive control for Ste6–166p and Sec61–2p.
