(A) MSS membranes from Ste13αHA-expressing vps21 strain BLY9 were combined with either MSS from kex2Δ strain (JBY209) or buffer on ice. The samples were immunoprecipitated with anti-HA after incubations at either 30 or 0°C. (B) MSS membranes from Kex2p- or Ste13αHA-expressing vps21 strain BLY9 (or JBY209 as VPS21 kex2Δ control) were combined as indicated under standard reaction conditions. Data are representative of three separate experiments performed using two different MSS preparations. (C) P200 membranes from JBY209 expressing Ste13αHA (VPS21 Ste13αHA) were combined with MSS membranes from JBY209 (pCWKX10) (VPS21 KEX2), BLY9 (pCWKX10) (vps21 KEX2), or BLY9 (vps21 kex2Δ) under standard reaction conditions. Reactions in B and C were performed in duplicate.
