Cytofluorographic analysis of KdFL binding to VV-infected L cells. The binding of KdFL1 and KdFL2 to L cells infected with rVVs expressing Kd, ER-retained Kd, or influenza virus hemagglutinin in the presence or absence of the indicated competitor peptides was determined by cytofluorography. The expression of Kd by rVV-infected cells also was monitored by indirect immunofluorescence using the anti-Kd mAb SF1.1.1. (42). Cell surface expression was determined using live cells. To enable detection of intracellular Kd, cells were fixed and permeabilized. This results in the loss of surface Kd, accounting for the decrease in mAb binding to VV-Kd-infected cells (13).
