Expression of stress PspA protein and stability of the CS YidC(C423R) and YidC(P431L) mutants at the nonpermissive temperatures. (A) Increased expression of the stress PspA protein at the nonpermissive temperature in the CS mutants. JS7131 bearing the CS YidC(C423R) or CS YidC(P431L) mutant on the pACYC184 vector was grown as described in Materials and Methods. Immunoblot analysis was performed to detect the PspA protein using polyclonal antibodies against PspA. The PspA levels were examined also with the arabinose-dependent JS7131 strain grown in either medium containing glucose to deplete YidC (Glu) or medium containing arabinose which expresses YidC at normal levels (Ara). (B) The CS YidC(C423R) and YidC(P431L) mutants are stable at the nonpermissive temperature. JS7131 bearing the CS YidC(C423R) or YidC(P431L) mutant on a pACYC184 vector was grown as described in Materials and Methods. As a control, YidC levels were also examined using the arabinose-dependent JS7131 strain grown in medium with glucose to deplete YidC (Glu) or arabinose with normal YidC levels (Ara).
