FMDV blocks NF-κB activity. (A) BHK-21 cells were cotransfected with a reporter plasmid expressing firefly luciferase driven by an NF-κB-dependent promoter (pNF-κB-LUC) and a control plasmid expressing Renilla luciferase constitutively (pRL-TK). Twenty-four hpi, cells were mock treated or infected with FMDV A12-IC (WT) or A12-LLV2 (leaderless) at different MOIs for the indicated times. A positive control with poly(I:C) was included. Relative luciferase activity (firefly/Renilla) was assayed on whole-cell lysates. RU, relative units. (B) Experiments were performed as described above (A), but 24 h prior to viral infection, poly(I:C) was transfected.
