Knockdown of USF1 leads to decreased methylation at H4R3 as well as acetylation of H3 and H4. USF1 expression was reduced 80 to 90% in 6C2 cells based on RNA interference technology (42). The results of ChIP assays with antibodies specific to dimethylated H4R3 (A), acetylated H3K9/K14 (B), and acetylated H4 (C) from wild-type (green) or USF1 knockdown (orange) 6C2 cell line are shown.
