(A) Immunoblot of SEK1 expression in control vector–transfected (pSilencer) or pSilencer-siSEK1 stably transfected (siSEK1#1 and siSEK1#2) single cell clones from DU-145 cells. Lysates containing equal amounts of protein (20 μg) were separated by SDS-PAGE and were immunoblotted with anti-SEK1 antibody. Control, untransfected cells. (B and C) Control plasmid or pSilencer-siSEK1 stably transfected cells were exposed to glucose-free medium for various times (10–120 min; B) or for 60 min (C). After incubating, cells were harvested, and Western blot analysis was performed with anti–ACTIVE JNK, anti-JNK2, or antiactin antibody as the loading control.
