Figure 2. — JNK-mediated phosphorylation of 14-3-3 results in the release of Bad from 14-3-3 in vitro. (a and b) GST-tagged proteins were subjected to an in vitro kinase assay with His-tagged MKK7-JNK and were incubated with His-tagged Bad wild type (WT; a) or Ser128A (b) that was preincubated with or without CA-Akt immune complex. The GST proteins were precipitated using glutathione–Sepharose beads. The amount of Bad protein coprecipitated with either GST or GST–14-3-3ζ was determined by immunoblot analysis (IB) with the indicated antibodies. The same results were obtained in at least two independent experiments.
