Figure 5. — Yotiao (AKAP9), PKA, and EGFR comprise a macromolecular signaling complex with IP₃R1. (A) Association of yotiao, IP₃R1, and EGFR with PKA in the DRM fraction. PC12 cell lysates were fractionated by sucrose density gradient centrifugation. DRM and soluble fractions were pooled and subjected to immunoprecipitation with an antibody against the catalytic subunit of PKA (PKA_Catα) or IgG. Precipitates were analyzed by blotting with the indicated antibodies. (B) Yotiao coimmunoprecipitates with IP₃R1, but not IP₃R3. PC12 cell lysates were immunoprecipitated with an antibody against IP₃R1 or IP₃R3. Precipitates were analyzed by blotting with anti-yotiao antibody. (C) PC12 cell lysates were immunoprecipitated with IgG or antibodies against EGFR. Precipitates were analyzed by blotting with the indicated antibodies. Primary antibody was omitted in lanes marked “−”. Immunoblots presented are representative of at least three independent experiments.
