After the addition of 25 μM MBC, at t0, MTs depolymerize to four short stubs (marked in red); one of them is connected to the SPB visualized by alp4-GFP. Two stubs disappear during the prolonged 10-min MBC incubation. After MBC washout, at t1, MT regrowth occurs from stubs (red arrowheads) and five new sites. (B) mto2Δ cell expressing alp4-GFP and GFP-tubulin (PT541). The bundle hardly shortens after MBC treatment, indicating a long stabilized region.
