Targeting of NCAM to lipid rafts is reduced in PrP / mice. (A) Brain homogenates or lipid rafts from brain homogenates and growth cones of PrP+/+ and PrP−/− mice (0–4 d old) were immunoblotted (WB) with antibodies against NCAM. The band at and above 180 kD represents polysialylated NCAM140 and NCAM180 (PSA-NCAM; Niethammer et al., 2002). In lipid rafts, a band representing nonpolysialylated NCAM120 is also observed. Diagrams show quantitation of PSA-NCAM and NCAM120. Note that the amount of PSA-NCAM is increased in brain homogenates and decreased in rafts isolated from brain homogenates and growth cones of PrP−/− mice. (B) Lipid rafts isolated from brain homogenates of PrP+/+ and PrP−/− mice (0–4 d old) were immunoblotted with L1 antibodies. Diagrams show quantitation of L1.
