HSL-GFP localizes to perilipin-coated lipid droplets in stimulated CHO cells. HSL-GFP was introduced transiently into CHO cells that were stably transfected to express perilipin A. Cells were stimulated for 10 min with 10 μM forskolin plus IBMX. The cells were fixed with 3% PFA and immunostained for perilipin, shown in red. HSL is shown in green. (A and B) Control cells without perilipin but with ADRP-coated droplet. (C–E) CHO cells stably transfected with native perilipin A. (F–H) CHO cells stably transfected to express perilipin A mutated in its three most NH2-terminal PKA sites. Red staining is Cy5 staining for perilipin; green is HSL-GFP. It is evident that HSL localized to the perilipin-coated droplets. Bar, 10 μm.
