(b) In vitro–translated ³⁵S-labeled GRASP65 proteins with the indicated aspartic acid to alanine substitutions were incubated with control or apoptotic HeLa cytosol in the absence or presence of 2 μM Ac-DEVD-CHO for 4 h at 37°C and analyzed by SDS-PAGE and autoradiography. (c) HeLa cells stably expressing COOH terminally GFP-tagged wild-type and D320A/D375A/D393A triple mutant forms of GRASP65 were incubated with 1 μM staurosporine for the times indicated. Cell extracts were analyzed by immunoblotting with antibodies to GFP and PARP.
