Figure 2. — Labeling nascent transcripts. (A) Approach. Wild-type (CHO-K1) cells are grown in [³H]uridine, and treated ± sarkosyl to extract completed transcripts and leave nascent ones. Polymerases (ovals) at different stages of the transcription cycle incorporate ³H (diamonds) into RNA (wavy lines), as some terminate and others initiate. Radioactivity in all transcripts increases progressively, whereas that in nascent ones plateaus. (B) Incorporation (expressed as a fraction of the total at 120 min).
