Folding of CTA1 inside the microsomes. CTA1 was translated for 10 min and the microsomes were isolated by centrifugation. One half was boiled for 5 min in 0.5% SDS, the other half was kept on ice. Both samples were diluted with 5 vol PBS containing 1% TX-100, divided into aliquots, incubated for 30 min on ice with the indicated concentrations of trypsin (upper panel) or proteinase K (lower panel), and analyzed by SDS-PAGE and autoradiography.
