HdmX stimulates Hdm2-mediated ubiquitination of p53. (A) Increasing amounts of bacterially expressed GST fusion proteins of Hdm2 and HdmX, respectively, were incubated with in vitro translated radiolabeled p53 under standard ubiquitination conditions for 2 h. The reaction products were analyzed by SDS/PAGE followed by fluorography. Amounts of Hdm2 and HdmX used are indicated in 10–1 μg. (B) Determination of the requirements for ubiquitination of in vitro translated p53 by bacterially expressed Hdm2 (250 ng) and by Hdm2/HdmX (25 ng each), respectively. E2, UbcH5.
