Calpain, but not caspase-8, is increased in HeLa-S3 cells, while calpain is not elevated in U2-OS cells, after exposure to cisplatin (CDDP). (a) Left panel: while there was no change after 15 min of incubation, after 1 h of exposure to 1 μM cisplatin, active calpain was highly increased in HeLa-S3 cells and this increase was blocked by 50 μM 2-APB (n=13). Right panel: this effect was not observed in U2-OS cells (n=6; 1 μM cisplatinfor 15 min and 1 h of exposure). (b) Cisplatin did not increase the large or small subunit of caspase-8 in HeLa-S3 cells after 1 h of exposure, while the activation of the TRAIL receptor showed that the caspase-8 antibody was effective (n=9). 2-APB, 2-aminoethoxydiphenyl borate; TRAIL, TNF-related apoptosis-inducing ligand.
