Analysis of Vα3.2-like TCR mutants. Mutants exhibited equivalent reactivity toward the Vβ8-specific mAb KJ16 (A). Vα3.2-like mutants were impaired in their ability to inhibit 30-5-7 (anti-L^d) Fab binding to cell surface QL9/L^d (B). Summary data indicate the relative change in binding free energy for mutant TCR compared with wild-type (C). Free energy changes were calculated from competition experiments using IC_50(mutant)/IC_{50(wild-type)}. Mutants with no detectable binding in the cell binding assay were assigned ΔΔG values = 1.5 kcal/mol, the sensitivity limit of the assay. ΔΔG values >0 indicate decreased binding.
