Susceptibility to antagonist ligands corresponds to the time of commitment. Vα2Vβ5 58CD8α/β cells were stimulated with P1.32K^b cells which had been prepulsed with 100 pM SIINFEKL peptide for 4 h at 37°C. At various time points after the initiation of the stimulation, antagonist peptides E1 or P7, control peptide K4, anti-K^b blocking antibody Y3, or medium alone were added, and the cells were resuspended. After 25 h of culture, the supernatant was harvested and assayed for IL-2 production. For more convenience, the kinetic analysis was carried out in two parts separated by 12 h. The mean of secreted IL-2 obtained in the samples where no antagonist peptides were added was set as 100%. The data are representative of four independent experiments.
