Response of LACK-specific transgenic T cells to altered LACK peptides. Spleen cells (106) from ABLE TCR-Cα0 mice were incubated with increasing amounts of the designated peptides for 48 h. Supernatants were collected from duplicate wells and assayed for IL-4 (center) and IFN-γ (right) by ELISA. Additional duplicate wells were pulsed with [3H]thymidine and incubated an additional 18 h before harvesting to assess proliferation by incorporation of radioactivity (left). No proliferation occurred in the absence of LACK peptide. Cells from TCR-Cα0 mice generated no signals in these assays (not shown).
