Reconstitution of lethally irradiated mice with purified bone marrow stem cells infected with recombinant Bcl-X_L retrovirus. Lethally irradiated BALB/c mice were reconstituted with stem cells purified from bone marrow of DO.11 WT or DO.11 CD28^−/− mice infected with control retrovirus or with retrovirus expressing Bcl-X_L. (A) Spleen and lymph nodes were harvested at 6–7 wk after transfer and analyzed for KJ1-26 and CD4 expression by flow cytometry. The BALB/c control was a nonirradiated age- and sex-matched mouse. Irradiated BALB/c mice that did not receive any bone marrow died within 10 d of treatment. (B) FACS^® analysis of GFP expression in CD4⁺KJ1-26⁺ cells from peripheral blood, spleen, and lymph nodes of BALB/c mice reconstituted with purified DO.11 CD28^−/− bone marrow stem cells infected with Bcl-X_L virus.
